Overview

NF-kappa-B inhibitor-like protein 1 (NFKBIL1) is a divergent member of the I-kappa-B family encoded by the NFKBIL1 gene, which lies within the major histocompatibility complex (MHC) class I region on chromosome 6[2][3][6]. Although initially thought to inhibit NF-kappa-B signaling as other I-kappa-B proteins do, functional studies show it does not bind or inhibit NF-kappa-B, but rather acts as a negative regulator of Toll-like receptor and interferon-regulatory factor signaling pathways, contributing to the negative regulation of transcriptional activation of NF-kappa-B target genes in response to proinflammatory stimuli[1][3][7]. NFKBIL1 is expressed in a tissue-specific manner and may also associate with mRNA, indicating a possible additional role in mRNA processing or regulation of translation[1]. Variants of this gene have been associated with susceptibility to rheumatoid arthritis and possibly other immune-mediated diseases[1][3]. At present, there are no clinically established interacting drugs or direct biomarker or safety data for therapeutic targeting of NFKBIL1.
