Overview

NFIA antisense RNA 1 (NFIA-AS1) is a long non-coding RNA found within intron 2 of the human NFIA gene, located on chromosome 1p31.3. It acts as a regulator of cholesterol homeostasis and inflammatory processes in vascular cells, particularly macrophage-derived foam cells and vascular smooth muscle cells. NFIA-AS1 modulates gene expression via “miRNA sponge” mechanisms, affecting miR-382-5p and miR-125a-3p activity. Elevated levels have been detected in atherosclerosis and some cancers, and knockdown reduces pathologic cell proliferation, inflammation, and adhesion responses in vascular models. Its biological pathways encompass lipid metabolism, cell death, and chronic inflammation, making it a promising candidate for therapeutic intervention in cardiovascular and malignant diseases. Key references supporting this information are peer-reviewed research articles and curated genetic databases. Specific drug interactions, clinical safety, and approved therapeutic agents are not yet established for NFIA-AS1, reflecting its classification as an emerging lncRNA target.

Mechanism of action

Mechanism involves modulation of miRNA axes [e.g., miR-382-5p/NFIA and miR-125a-3p/AKT1], but no approved drugs act on this pathway directly.

03

Biological functions

Modulation of cholesterol homeostasisRegulation of inflammatory responseRegulation of cell proliferation and apoptosisFoam cell formationRegulation of vascular smooth muscle cell (VSMC) biology

Disease associations

AtherosclerosisCardiovascular diseaseCancer, including breast cancer, high grade glioma, gastric cancer
05

Safety considerations

No safety profile is established; targeting lncRNAs presents general challenges such as off-target effects and delivery efficacy.

Biomarkers

Increased NFIA-AS1 expression in serum/tissue is correlated with atherosclerosis and certain cancers
