NFS1 cysteine desulfurase is an essential, pyridoxal phosphate (PLP)-dependent enzyme primarily involved in the biosynthesis of iron–sulfur (Fe-S) clusters. These clusters are critical cofactors for a wide range of cellular enzymes and processes. The enzyme catalyzes the removal of sulfur from L-cysteine, producing L-alanine and a persulfide intermediate, which donates sulfur for Fe-S cluster assembly. It operates as part of a multi-protein complex that includes ISD11, ACP (acyl carrier protein), ISCU2 (scaffold protein), and FXN (frataxin). High expression levels are necessary under physiological oxygen conditions and suppression leads to growth inhibition in certain tumor types. Phosphorylation status can influence chemosensitivity to drugs like oxaliplatin.
