Overview

NGcGM3 ganglioside is a glycosphingolipid expressed in various human cancers but absent in normal human tissues. Its selective expression makes it a valuable target for cancer immunotherapy approaches, including antibodies, vaccines, and CAR T-cell therapies. It is considered a tumor-associated antigen and a negative prognostic marker in several cancers.

Mechanism of action

Racotumomab and mAb 14F7 bind to NGcGM3 on tumor cells, leading to antibody-dependent cell-mediated cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), or direct induction of apoptosis. CAR T cells engineered to target NGcGM3 directly kill tumor cells.
