NGFI-A binding protein 1 (NAB1) is a transcriptional corepressor that interacts primarily with zinc finger transcription factors from the early growth response (EGR) family, such as EGR1 and EGR2, to repress their ability to activate transcription. NAB1 is localized in the nucleus and exerts its corepressor function by binding to the R1 domain of EGR-family proteins when they are DNA-bound, without interfering with DNA binding itself. This direct repression mechanism modulates gene expression related to development, immune responses, and neural function. NAB1 also influences diverse biological programs, such as endochondral ossification, nervous system development, and regulation of inflammation by silencing genes like IFNGR1, thereby dampening interferon-mediated responses. In addition, the NAB1 locus produces a circular RNA species called circ-NAB1, which has been implicated as an oncogenic regulator in endometrial cancer by modulating cell proliferation, migration, and the cell cycle. NAB1, due to its indirect regulatory nature and absence of known drug interactions, is not currently regarded as a direct therapeutic target or receptor. However, its regulatory RNAs and interaction networks make it of interest in disease-related research, particularly cancer and immune modulation.
