Overview

NGFI-A binding protein 2 (NAB2) is a nuclear protein acting primarily as a transcriptional corepressor of EGR (early growth response) family transcription factors, notably EGR1 and EGR2, thus regulating their gene activation in a negative feedback manner[1][2][7]. NAB2 functions through direct protein-protein interactions and recruitment of chromatin remodeling complexes such as NuRD, influencing processes like angiogenesis, cellular differentiation, and tissue repair[1][2]. Pathologically, NAB2 is notable for its role in gene fusions with STAT6 in solitary fibrous tumors and hemangiopericytomas, altering its function from transcriptional repressor to activator and serving as a molecular hallmark of these tumors[2]. There are no known drugs approved to target NAB2 directly, nor established safety concerns, but the NAB2-STAT6 fusion is used as a diagnostic biomarker for relevant tumor types[2]. NAB2 is not classified as a classical therapeutic target (such as a receptor, enzyme, ion channel, or transporter), but rather as a transcription regulator/corepressor.
