Overview

NHS-like protein 2 (NHSL2) is a cytoplasmic protein predicted to have calcium ion binding activity and to be involved in cell differentiation[1][3][5][9][11]. The gene encoding NHSL2 (HGNC: 33737; NCBI Gene: 340527) is a paralog of the NHS gene. NHSL2 spans 1,189 amino acids and contains a WAVE homology domain (WHD), suggesting a possible role in regulating cytoskeletal dynamics[3]. Expression data indicate cytoplasmic presence in several human tissues, including the cerebellum[9]. The protein is primarily predicted from sequence and in silico analysis rather than extensive functional studies, and there are currently no drugs, targeted therapies, or biomarker applications associated with NHSL2[1][3][5][11]. No evidence supports its direct involvement in major disease processes outside rare genetic associations (e.g., Nance-Horan syndrome)[1]. The alternative alias "TPT1-like protein" refers to sequence similarity, but NHSL2 should not be confused with TPT1 (Translationally Controlled Tumor Protein), which has more substantial roles in apoptosis, cancer, and immune regulation[2][6][8][10].
