NHS-like protein 3 (NHSL3), also known as KIAA1522, is a member of the NHS family proteins and functions as a regulator of single and collective cell migration[1][3]. It displays several isoforms, including a unique short isoform that specifically regulates migration persistence in single cells[1]. NHSL3 can interact with proteins involved in actin dynamics, such as the WAVE complex (notably in long isoforms), but regulates migration persistence via mechanisms distinct from other NHS family members like NHSL1[1]. Additionally, NHSL3 is predicted to activate the TNF-NFκB signaling pathway and participate in cell differentiation[3][4][6]. Its dysregulation has been linked to cancer progression, particularly in breast cancer, and gene expression data suggest involvement in some corneal pathologies[3][5]. There are currently no approved drugs that specifically target NHSL3, and its utility as a biomarker or therapeutic target remains unestablished[3][5].
