Overview

Niban apoptosis regulator 1 (NIBAN1) is a protein encoded by the NIBAN1 gene (formerly FAM129A/C1orf24) on chromosome 1q25.3. It belongs to the family with sequence similarity 129 (FAM129), sharing a pleckstrin homology (PH) domain in its N-terminal region. NIBAN1 is highly expressed in multiple cancer types and some normal tissues, especially under stress conditions, where it participates in regulating key processes such as apoptosis (including p53-mediated events), cell proliferation, survival under cellular stress, and autophagy. NIBAN1 is implicated in several cancer types and is being explored as a diagnostic and/or prognostic biomarker, particularly because its expression increases during early carcinogenesis in tissues like kidney and thyroid. Though it is often referred to as a "regulator" in biological stress responses, NIBAN1 is not a classical drug target such as a receptor, enzyme, or transporter, and there are currently no known drugs that directly target it[1][2][3][4].
