Nicotinamide adenine dinucleotide (NAD) biosynthesis refers to the cellular pathways that produce NAD, a vital coenzyme found in all living cells. NAD exists in two forms: oxidized (NAD⁺) and reduced (NADH). It is essential for redox reactions, energy metabolism, DNA repair, and cell signaling. The main pathways for NAD biosynthesis are the de novo pathway (from tryptophan), the Preiss–Handler pathway (from nicotinic acid), and the salvage pathway (from nicotinamide). Supplementation with NAD+ precursors is being explored to improve metabolic health and address age-related decline.
