Overview

The neutrophil superoxide anion generation pathway is a fundamental mechanism of the innate immune system, primarily executed by the Nicotinamide adenine dinucleotide phosphate (NADPH) oxidase complex, specifically the NOX2 isoform (UniProt: P04839). This enzyme complex is composed of membrane-bound subunits, including Cytochrome b-245 heavy chain (gp91phox) and light chain (p22phox), and several cytosolic regulatory subunits such as p47phox, p67phox, and p40phox (PubMed: 22226317). Upon activation by inflammatory stimuli, these components assemble at the membrane to transfer electrons from NADPH to molecular oxygen, producing superoxide anions (O2•−) in a process known as the respiratory burst (PubMed: 15013790). This pathway is essential for the destruction of phagocytosed pathogens, and its genetic deficiency leads to Chronic Granulomatous Disease (CGD), characterized by severe recurrent infections (StatPearls: NBK537072). Conversely, excessive superoxide production is a major driver of oxidative stress and tissue damage in chronic inflammatory diseases, cardiovascular disorders, and neurodegeneration (PubMed: 28249116). Therapeutic targeting of this pathway involves small molecule inhibitors like GSK2795039, which aim to reduce pathological ROS levels while ideally preserving sufficient immune function (PubChem: CID 118971114).
