Overview

Nicotinic acetylcholine receptors (nAChRs) are pentameric ligand-gated ion channels that mediate fast synaptic transmission in the central and peripheral nervous systems, playing key roles in neuromuscular signaling and cognitive processes (StatPearls, 2023). AMPA receptors (α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors) are tetrameric ionotropic glutamate receptors that facilitate the majority of fast excitatory neurotransmission in the brain and are essential for synaptic plasticity and memory formation (IUPHAR/BPS, 2024). GABA receptors, primarily the ionotropic GABA-A subtype, are pentameric chloride-selective channels that serve as the primary mediators of inhibitory neurotransmission in the mammalian central nervous system (NCBI, 2022). These three receptor systems collectively maintain the excitatory-inhibitory (E/I) balance required for normal brain function; their dysregulation is central to the pathophysiology of epilepsy, Alzheimer's disease, schizophrenia, and various anxiety disorders (PubMed, 2023). Pharmacological intervention targeting these receptors is diverse, involving agonists like nicotine for nAChRs, non-competitive antagonists like perampanel for AMPA receptors, and positive allosteric modulators like benzodiazepines for GABA-A receptors (PubChem, 2024). Given their widespread expression and critical roles, drugs affecting these targets often require careful dosing to avoid significant side effects such as sedation, respiratory depression, or cognitive impairment.
