The α3β4 nicotinic acetylcholine receptor (nAChR) is a pentameric ligand-gated ion channel primarily found in autonomic ganglia, the adrenal medulla, and specific brain regions like the medial habenula and interpeduncular nucleus. It plays a crucial role in fast synaptic transmission by mediating cation influx upon activation by acetylcholine or other agonists. This receptor subtype is implicated in drug-seeking behavior, nicotine reward, and physical withdrawal symptoms. Genetic variations in the CHRNA5/A3/B4 gene cluster, which encodes the α3 and β4 subunits, are associated with increased risk for nicotine dependence and other diseases. Selective antagonists of the α3β4 nAChR are being investigated as potential anti-addictive therapies.
