Overview

The nicotinic acetylcholine receptor alpha4 beta2 (α4β2 nAChR) is a neuronal ligand-gated ion channel composed of α4 and β2 subunits. It is the most abundant nicotinic receptor subtype in the brain and plays a critical role in synaptic transmission, learning, attention, and addiction. This receptor mediates fast synaptic transmission by increasing membrane permeability to sodium (Na+) and potassium (K+) ions upon activation by acetylcholine or nicotine. Mutations in CHRNA4 gene encoding α4 can lead to short stature or predispose individuals toward obesity due to altered growth hormone regulation.

Agonist binding opens the ion channel, allowing Na+ influx and K+ efflux, leading to neuronal depolarization and excitation.
