Overview

Nidogen-1 (NID1) is a highly conserved sulfated glycoprotein and critical extracellular matrix component of basement membranes in nearly all tissues[1][3][4][9]. It serves as a structural linker that tightly binds laminin, collagen IV, perlecan, and fibronectin, bridging the networks formed by these molecules to maintain basement membrane stability and integrity[1][4][9]. Nidogen-1 supports tissue architecture, influences cell adhesion and migration, and regulates developmental processes and the stem cell niche[3][4]. Mutations or dysregulation of NID1 are implicated in developmental syndromes such as Dandy–Walker malformation and have been associated with tumor progression, particularly in cancers of epithelial origin[1][4][7][9]. While essential for basement membrane structure and function, there are currently no known drugs that directly modulate nidogen-1, nor is it a typical therapeutic target in clinical pharmacology.
