The Nidovirus RdRP-associated nucleotidyltransferase (NiRAN) is a unique enzymatic module universally present at the N terminus of all known nidoviral polymerases. Its essential role in nucleotide transfer reactions required for efficient genome replication, transcription initiation, and possibly mRNA capping makes it both fundamental to virus biology and an attractive target for broad-spectrum antivirals targeting diseases such as COVID-19.
