Overview

Ninein-like protein (NINL), also known as KIAA0980 and NLP, is a human centrosomal protein encoded by the NINL gene located on chromosome 20[3][5][6]. NINL is involved in microtubule organization, particularly nucleation and anchoring at the centrosome, and contributes to vesicle transport in photoreceptor cells[1][7]. It interacts with the γ-tubulin ring complex (γTuRC) and is regulated by cell cycle kinases such as Plk1 and Nek2, which control its phosphorylation and association with the centrosome[2][4]. NINL is implicated in the organization of the Golgi apparatus through its association with the dynein-dynactin complex[2]. Overexpression of NINL is observed in various human cancers, especially breast and lung carcinoma, where its gene amplification correlates with tumorigenic processes and genomic instability[4]. Variants in NINL are also associated with Heimler syndrome 2 and accommodative spasm[1]. While NINL is considered a promising target for research in cancer diagnostics and therapeutics, no direct drug interactions or established clinical inhibitors exist to date.
