The NineTeen Complex (NTC) is a multi-protein complex that plays an essential and conserved role in the regulation of pre-mRNA splicing, a critical step in eukaryotic gene expression. The NTC associates with the spliceosome—the large ribonucleoprotein machinery responsible for removing introns from pre-mRNA and ligating exons to form mature mRNAs. The NTC is required for both catalytic steps of intron removal during pre-mRNA splicing and regulates the formation, activation, and progression of key spliceosome conformations necessary for accurate and efficient splicing. During spliceosome activation, the NTC promotes specific RNA-RNA interactions that form the catalytic core needed for the first step of splicing. Proper function ensures fidelity in exon-intron recognition; defects can lead to aberrant mRNAs implicated in disease states including certain cancers and genetic disorders linked to mis-spliced transcripts.
