Overview

NIPBL divergent transcript (NIPBL-DT, also known as NIPBL-AS or NIPBL-AS1) is a long non-coding RNA (lncRNA) transcribed from a bidirectional promoter shared with the NIPBL gene[3][5]. The transcript is approximately 5.3 kb in length and does not code for a protein. Its primary function is transcriptional interference; when NIPBL-DT is transcribed, NIPBL gene transcription is reduced, and vice versa[3]. This mutual regulation likely serves as a fine-tuning mechanism to ensure appropriate levels of NIPBL, a gene critical for sister chromatid cohesion and gene regulation during development. Altered expression of NIPBL-DT and subsequent dysregulation of NIPBL abundance is implicated as a contributing factor in Cornelia de Lange syndrome (CdLS)[3][6]. NIPBL-DT itself is not a direct drug or small molecule target, nor is it a conventional receptor, enzyme, or transporter, but its role in NIPBL gene regulation is of clinical significance for genetic syndromes such as CdLS.
