Overview

Nitric oxide synthases (NOS) are a family of enzymes that catalyze the production of nitric oxide (NO) from L-arginine. NO is a critical signaling molecule involved in numerous physiological processes, including neurotransmission, vascular regulation, and immune response. There are three main isoforms: NOS1 (neuronal), NOS2 (inducible), and NOS3 (endothelial). Dysregulation of NOS activity is implicated in various diseases.

Mechanism of action

NOS inhibitors block the production of nitric oxide by inhibiting the enzymatic activity of NOS isoforms. This can involve competitive binding to the active site, disruption of cofactor binding, or interference with the enzyme's dimerization or conformational changes.
