Overview

Neuronal nitric oxide synthase (nNOS), also known as NOS I, is an enzyme responsible for the synthesis of nitric oxide (NO) in neurons. NO acts as a gaseous signaling molecule with diverse roles in the central and peripheral nervous systems. nNOS activity is calcium/calmodulin-dependent; it catalyzes the oxidation of L-arginine to produce NO and citrulline when activated by increased intracellular Ca²⁺ levels following NMDA receptor stimulation. Altered nNOS function has been implicated in various neurological disorders due to its role in synaptic signaling, neurodevelopmental processes, cardiovascular regulation, oxidative stress response in myocardium, heart failure prevention, basal blood flow maintenance, and autonomic cardiac control.
