Nitric oxide synthase 1 (NOS1) mRNA is the messenger RNA transcript that encodes the neuronal isoform of the enzyme nitric oxide synthase (nNOS). This enzyme is responsible for the production of nitric oxide (NO), a critical gaseous signaling molecule involved in neurotransmission, synaptic plasticity, and the regulation of smooth muscle tone in the gastrointestinal and respiratory tracts (UniProt P29475; NCBI Gene ID: 4842). Dysregulation of NOS1 mRNA expression is implicated in several pathological states, including neurodegenerative disorders like Alzheimer's and Parkinson's diseases, where excessive NO production contributes to oxidative stress and neuronal death (Zhou & Zhu, 2009). Additionally, altered NOS1 mRNA levels are linked to chronic pain conditions and psychiatric disorders such as schizophrenia. Targeting the mRNA transcript using antisense oligonucleotides (ASOs) or RNA interference (RNAi) offers a highly specific therapeutic approach to downregulate nNOS expression without affecting other isoforms like endothelial (eNOS) or inducible (iNOS) nitric oxide synthase. Preclinical studies have explored these RNA-targeted strategies to mitigate neurotoxicity and hyperalgesia in various disease models (Ishii et al., 2002).
