Nitroreductase-1 (NTR1)

Target
NTR1
Molecular classification
Enzyme, Flavoprotein
01

Overview

Nitroreductase-1 is a member of the nitroreductase family of enzymes that catalyze the reduction of nitro-containing compounds, particularly nitroaromatics. It is a flavoprotein that utilizes flavin mononucleotide (FMN) as a cofactor and typically functions as a homodimer. Nitroreductase-1 plays roles in bacterial metabolism and is used in cancer gene therapy to convert non-toxic prodrugs into cytotoxic agents under hypoxic conditions typical of tumor microenvironments. It shows broad substrate specificity and operates via a ping-pong bi-bi mechanism.

Mechanism of action

Catalyzes two-electron reductions of nitroaromatic compounds to their corresponding nitroso derivatives and further to hydroxylamines, utilizing NADH or NADPH as electron donors. Operates via a ping-pong bi-bi mechanism.
