Overview

Nitroxyl donors are compounds that release nitroxyl (HNO), the one-electron reduced and protonated form of nitric oxide (NO). HNO is chemically distinct from NO, exhibiting unique biological activities. These donors have therapeutic potential in cardiovascular diseases like heart failure and hypertension, acting primarily through direct post-translational modification of thiol residues on target proteins. Examples include Angeli’s salt, Piloty’s acid, acyloxy nitroso compounds, BMS‐986231, and CXL-1020.

Mechanism of action

Releases nitroxyl (HNO) which modifies protein thiols, leading to vasodilation and improved cardiac function. Specifically, HNO reacts with cysteine residues to form N-hydroxysulfenamide intermediates and induces interdisulfide formation between specific cysteines on PKGIα kinase monomers, mimicking cGMP binding and activating PKGI without requiring cGMP itself.
