NK2 homeobox 8 (NKX2-8) is a homeobox-containing transcription factor within the NKX2 family, highly conserved in evolution. It plays a crucial role in the development and differentiation of several organs, especially the lung, thyroid, pharynx, spinal cord, and hindbrain. In normal physiology, NKX2-8 regulates genes involved in cell lineage specification and cell proliferation, particularly in tracheobronchial stem cells and neural tissue[1][3][5]. Alterations in NKX2-8 are associated with neural tube defects and various cancers, including lung, esophageal, hepatic, ovarian, and urothelial carcinoma. Its role in cancer appears context-dependent, functioning as an oncogene in some cases and a potential tumor suppressor in others. NKX2-8 is also used as a biomarker for prognosis in certain cancers, with its expression levels correlated to disease progression and drug resistance, particularly to platinum-based chemotherapy such as cisplatin[1][5]. Molecularly, NKX2-8 acts via DNA binding and interactions with transcriptional machinery, affecting apoptosis and fatty acid oxidation pathways among others[1][3]. There are no specific approved drugs targeting NKX2-8, but its expression profile is clinically significant for cancer prognosis and therapeutic resistance.
