Overview

NK6 homeobox 3 (NKX6.3) is a transcription factor belonging to the NKX6 subfamily of homeodomain proteins, primarily involved in the development and differentiation of the central nervous system, gastrointestinal tract, and pancreas[5][7]. In adult tissues, NKX6.3 is particularly important as a regulator of gastric epithelial cell differentiation, where it upregulates markers of gastric lineage (such as SOX2, Muc5ac, and GKN1) and represses intestinal markers (such as CDX2 and Muc2)[1][4]. NKX6.3 functions as a tumor suppressor in the stomach by arresting cell proliferation, promoting apoptosis (via death receptor and mitochondrial pathways), and inhibiting cancer cell migration, invasion, and epithelial-to-mesenchymal transition through transcriptional repression of genes involved in the Wnt/β-catenin and Rho-GTPase pathways[1][2][4]. Loss or reduced expression of NKX6.3 is associated with poor prognosis, metastasis, and genomic instability in gastric cancer, due in part to its role in DNA replication and repair regulation[3][4]. No interacting drugs or specific mechanisms of therapeutic action have been reported as of the latest data, but reduced NKX6.3 expression is being explored as a potential biomarker for aggressive gastric cancer and therapeutic stratification[2][3][4].
