Overview

NKG2-C (also known as NKG2C) is an activating receptor expressed on human natural killer (NK) cells, belonging to the NKG2 family (CD159) within the C-type lectin-like receptor superfamily. It forms a heterodimer with CD94 and associates with the adaptor protein DAP-12 through a positively charged residue in its transmembrane region, lacking ITIMs but enabling ITAM-based signaling that promotes NK cell cytotoxicity, degranulation, and cytokine production upon binding HLA-E ligands. Unlike inhibitory NKG2A/B, NKG2-C activates NK cells, contributing to immune surveillance against tumors and infected cells, with higher expression in adaptive NK cells relevant to graft-versus-tumor effects.
