NKG2D is a type II transmembrane glycoprotein that functions as an activating and costimulatory receptor in the immune system. It is primarily expressed on natural killer (NK) cells, but also found on subsets of T cells. It recognizes stress-induced self-proteins that are structurally related to MHC class I molecules, playing a key role in immunosurveillance against tumors and virus-infected or stressed/damaged self-cells. The NKG2D-ligand axis is implicated in cancer immunosurveillance, and NKG2D pathway modulation has therapeutic potential, especially in cancer immunotherapy.
