Overview

NKX2-1 antisense RNA 1 (NKX2-1-AS1) is a long non-coding RNA (lncRNA) located in chromosomal region 14q13.3, overlapping the protein-coding gene NKX2-1[1][7]. It does not encode a protein, but instead regulates gene expression independently of NKX2-1[1]. NKX2-1-AS1 is highly expressed in lung adenocarcinomas versus non-tumor or squamous cell controls[1]. Functionally, NKX2-1-AS1 modulates the expression of genes related to cell adhesion, migration, and immune checkpoints, notably negatively regulating CD274/PD-L1, thereby potentially influencing immune evasion in cancer[1]. It has also been implicated in promoting tumor progression and angiogenesis in gastric cancer, specifically via upregulation of SERPINE1 and activation of the VEGFR-2 pathway[6][7]. NKX2-1-AS1 may act as a microRNA sponge and is more stable than the NKX2-1 mRNA[4][1]. Although its biological roles are actively studied, it is not considered a canonical therapeutic target such as a receptor, enzyme, or transporter, and no drugs are known to specifically target NKX2-1-AS1 at present.
