Overview

NLGN1 antisense RNA 1 (NLGN1-AS1) is a cytoplasmic long non-coding RNA found to be upregulated in clear cell renal cell carcinoma. It mainly functions by sponging miR-136-5p, leading to increased expression of FZD4 and activation of the oncogenic Wnt/β-catenin signaling pathway, thereby promoting cancer cell proliferation. Knockdown of NLGN1-AS1 results in reduced proliferation and inhibition of this pathway, suggesting that NLGN1-AS1 may serve as a novel therapeutic target in cancers such as ccRCC. Its role is supported by both bioinformatic and experimental studies, but there are currently no approved drugs targeting NLGN1-AS1 directly[3][6].

Mechanism of action

Knockdown (gene silencing) of NLGN1-AS1 decreases cell proliferation via upregulation of miR-136-5p and inhibition of the Wnt/β-catenin signaling pathway. Acts by sponging and regulating specific miRNAs (e.g., miR-136-5p) which in turn affect downstream target proteins like FZD4.
