Overview

NLR family CARD domain-containing protein 3 (NLRC3) is a cytosolic pattern recognition receptor in the NOD-like receptor family. NLRC3 contains a CARD domain, a nucleotide-binding (NBD) domain, and leucine-rich repeats (LRR), and is primarily expressed in immune cells. Unlike other NOD-like receptors that act as positive innate immune sensors, NLRC3 serves as a negative regulator, attenuating activation of NF-κB and STING pathways, suppressing inflammasome formation, and limiting cell proliferation. It prevents inflammasome assembly by interacting with ASC and pro-caspase-1, modulates T cell activation and differentiation, and negatively regulates the PI3K-AKT-mTOR pathway, thus controlling cell fate and immune responses. NLRC3 is implicated in a range of immune-mediated diseases, including infections, sterile inflammation, and cancer, largely through its roles in dampening immune responses and cell proliferation[1][2][3].
