NLRP3 inflammasome activation is a key event in the innate immune system, triggered by various stimuli including PAMPs, DAMPs, and cellular stress. This leads to the assembly of a multiprotein complex (NLRP3, ASC, Caspase-1) and the subsequent activation of caspase-1. Active caspase-1 cleaves pro-IL-1β and pro-IL-18 into their mature forms, leading to their secretion and promoting inflammation. Aberrant activation is linked to several diseases, making it a therapeutic target for inflammatory disorders.
