NMDA receptor synaptonuclear signaling and neuronal migration factor (NSMF, formerly known as NELF or Nasal embryonic LHRH factor) is a protein encoded by the NSMF gene, which is highly conserved across mammals[1][2][5]. This protein is crucial for the guidance of olfactory axon projections and the migration of gonadotropin-releasing hormone (GnRH; also known as luteinizing hormone-releasing hormone, LHRH) neurons during embryonic development[1][5]. NSMF couples NMDA receptor (a type of glutamate receptor) signaling at synapses to the nucleus and participates in the regulation of long-lasting changes in neuronal gene expression, specifically through modulating CREB (cAMP response element-binding protein) signaling pathways[3][5]. NSMF functions as a synapto-nuclear messenger: following NMDAR activation, it is transported to the nucleus where it can affect gene transcription and neuronal plasticity[3]. Disruption or mutations in NSMF are strongly associated with idiopathic hypogonadotropic hypogonadism (IHH) and Kallmann syndrome, disorders marked by defective puberty and infertility frequently accompanied by anosmia or hyposmia (impaired sense of smell)[2][5]. NSMF knockout in model organisms impairs neuronal migration and hippocampal function, leading to neurological and behavioral abnormalities[3]. Although the protein has features of a transcriptional regulator and is sometimes categorized as a transcription factor, its principal recognized molecular role is as a synapto-nuclear signaling molecule essential for neurodevelopment and neuronal gene regulation[3][5]. There are no known drugs that directly target NSMF, and it is not widely regarded as a therapeutic target, but its mutations serve as a biomarker for diagnosis of certain hypogonadotropic conditions[2][5]. NSMF is a synapto-nuclear signaling molecule important for the embryonic migration of neuroendocrine and olfactory neurons, regulation of neuronal gene expression, and synaptic plasticity, with dysfunction causing specific neurodevelopmental disorders[1][2][3][5].
