The classification "No conventional drug-like molecular target" refers to therapeutic agents that do not exert their pharmacological effects through interaction with a specific biological macromolecule, such as a receptor, enzyme, or ion channel. Instead, these agents typically function through non-specific physical or chemical mechanisms (ChEMBL, 2024). For example, antacids like calcium carbonate work by directly neutralizing gastric acid, while osmotic laxatives like magnesium sulfate create an osmotic gradient to retain water in the intestinal lumen (Goodman & Gilman's, 2023). Other examples include activated charcoal, which adsorbs toxins in the gastrointestinal tract, and simethicone, which alters the surface tension of gas bubbles to facilitate their expulsion. Because these substances lack a defined protein binding site, they do not follow traditional receptor-mediated pharmacodynamics and are often excluded from standard target-based drug discovery efforts. Safety concerns associated with these agents often involve systemic electrolyte disturbances or the non-specific interference with the absorption of other co-administered medications.
