Overview

The classification "No conventional molecular target" refers to a group of therapeutic agents that do not exert their effects by binding to specific biological macromolecules such as receptors, enzymes, or transporters (DrugBank, 2024). Instead, these substances function through non-specific physical or chemical mechanisms within the body. For example, antacids like magnesium hydroxide work by directly neutralizing gastric acid through a simple chemical reaction (StatPearls, 2023). Osmotic agents, such as mannitol or polyethylene glycol, create physical osmotic gradients to shift water between physiological compartments, which is useful in treating edema or constipation (PubMed, PMC7465231). Adsorbents like activated charcoal physically trap toxins and chemicals in the gastrointestinal tract to prevent systemic absorption (NIH, 2023). Additionally, agents like sugammadex act by encapsulating other drug molecules rather than interacting with a human protein target (FDA, 2015). While these agents often have a broad safety profile, they can lead to complications such as electrolyte imbalances or the unintended malabsorption of other medications.
