Overview

The term No conventional pharmacological target refers to a classification for drugs that do not interact with specific molecular structures like receptors or enzymes to produce a therapeutic effect (DrugBank, 2023). Instead, these agents act through broad physical or chemical mechanisms, such as altering the pH of a biological fluid or changing the osmotic pressure within a compartment (Goodman & Gilman, 2018). For instance, antacids like aluminum hydroxide work by directly neutralizing hydrochloric acid in the stomach, while osmotic laxatives like polyethylene glycol retain water in the intestinal lumen to facilitate bowel movements (StatPearls, 2023). Other examples include activated charcoal, which uses its high surface area to adsorb toxins and prevent their absorption into the bloodstream (PubMed, 2022). Because these substances lack a specific protein target, they often require much higher molar concentrations than targeted therapies and are less likely to be affected by genetic polymorphisms in target proteins. However, they can still cause significant systemic effects, such as electrolyte imbalances or interference with the absorption of other medications (NIH, 2023). This category is essential for distinguishing non-specific chemical therapies from the majority of modern drugs that rely on high-affinity molecular recognition.
