Nociceptor potassium channels are a diverse group of potassium (K⁺) channels expressed in nociceptive sensory neurons. These channels play a critical role in regulating neuronal excitability, stabilizing the resting membrane potential, and controlling the firing threshold of nociceptors—the primary sensory neurons responsible for detecting noxious (painful) stimuli. Downregulation or dysfunction of these K⁺ channels is associated with increased risk for chronic pain conditions. Because they limit neuronal excitability, enhancing K⁺ channel function is a promising strategy for novel analgesics. Relevant subtypes include voltage-gated Kv channels (e.g., Kv7) and two-pore domain K2P channels (e.g., TRESK/TREK).
