Overview

NOD-like receptor family pyrin domain containing 3 (NLRP3) is a cytosolic sensor protein that serves as the central component of the NLRP3 inflammasome, a multiprotein complex crucial for innate immune defense and inflammatory signaling[1][4][5][6]. Upon recognition of diverse pathogen- or damage-associated molecular patterns, NLRP3 oligomerizes via its NACHT domain, recruiting the adapter ASC (apoptosis-associated speck-like protein containing a caspase recruitment domain) through pyrin domain (PYD) interactions[1][3][4]. ASC then recruits and activates pro-caspase-1, resulting in its autocatalytic activation. Active caspase-1 processes the pro-inflammatory cytokines IL-1β and IL-18, and cleaves gasdermin D to induce pyroptosis[1][3][6]. NLRP3 activation contributes centrally to both host defense and the pathogenesis of autoimmune, autoinflammatory, and degenerative disorders[6][9]. Pharmacological targeting of NLRP3 is a major strategy under investigation for inflammatory and autoimmune diseases[6].
