Overview

The NLRP3 inflammasome pathway refers to a cellular signaling cascade initiated by the multiprotein NLRP3 inflammasome complex, which is a key sensor in the innate immune system detecting a wide array of exogenous and endogenous danger signals. Upon activation, NLRP3 nucleates the recruitment of the adaptor ASC and pro-caspase-1, ultimately leading to the activation of caspase-1, cleavage, and release of pro-inflammatory cytokines IL-1β and IL-18, and induction of pyroptotic cell death. Tight regulation is necessary as its dysregulation is implicated in many chronic inflammatory and metabolic diseases, neurodegeneration, cardiovascular pathologies, drug-induced toxicities, and autoinflammatory syndromes. The pathway is regarded as a significant therapeutic target, with small molecule inhibitors such as MCC950 and CY-09 demonstrating promising effects in preclinical models of inflammatory disease[1][2][4][5][6][7][8].
