Nod-like receptor family pyrin domain-containing protein 3 (NLRP3), also known as cryopyrin, is a key intracellular sensor in the innate immune system. It functions as a pattern recognition receptor (PRR) that detects pathogen-associated molecular patterns (PAMPs) and danger-associated molecular patterns (DAMPs). Upon activation, NLRP3 undergoes conformational changes leading to oligomerization, which recruits ASC and caspase-1 to form the inflammasome complex. Activated caspase-1 then processes pro-inflammatory cytokines IL-1β and IL-18 into their mature forms and cleaves gasdermin D to induce pyroptosis. Mutations or dysregulation in NLRP3 are associated with several autoinflammatory disorders, making it a therapeutic target for inflammatory diseases.
