Overview

NOD-like receptor pyrin domain containing 3 (NLRP3) is a cytosolic pattern recognition receptor that plays a central role in the innate immune system. It acts as a sensor component of the inflammasome, triggering inflammation and pyroptosis upon activation by various stimuli. Mutations or dysregulation are linked to autoinflammatory diseases and implicated in chronic inflammatory conditions.

Mechanism of action

NLRP3 acts as a sensor component of the inflammasome. Upon activation by PAMPs and DAMPs, it oligomerizes and recruits adaptor proteins (ASC), activating caspase-1 which processes pro-inflammatory cytokines (IL-1β and IL-18) into their mature forms and induces pyroptosis.
