Nogo-A, also known as neurite outgrowth inhibitor A, is a member of the reticulon family (RTN4). It is a potent myelin-associated inhibitor of neurite outgrowth in the central nervous system (CNS), limiting axonal regeneration and plasticity after CNS injury. Nogo-A functions by binding to neuronal receptors (NgR1 complex), triggering intracellular signaling cascades involving RhoA, and destabilizing actin filaments at growth cones, halting further extension. Blocking antibodies against Nogo-A have shown promise in promoting axonal regeneration and compensatory sprouting following CNS lesions in animal models.
