Overview

Non-coding repressor of NFAT (NRON) is a long non-coding RNA (lncRNA) that negatively regulates the activity of the transcription factor NFAT by sequestering it in the cytoplasm within a multiprotein complex, thereby limiting its nuclear translocation and transcriptional activity. NRON functions as a scaffold that assembles regulatory proteins, including IQGAP1, KPNB1, and LRRK2, to retain phosphorylated NFAT outside the nucleus in resting cells. Upon stimulation or NRON knockdown, increased NFAT nuclear translocation occurs, enhancing NFAT-dependent gene expression such as cytokine production. NRON has been implicated in modulation of HIV replication by regulating NFAT activity in CD4 T cells, and altered NRON expression is associated with diseases like hepatocellular carcinoma and atrial fibrosis. NRON is not considered a conventional drug target (e.g., receptor, enzyme) at this time, but it is a key regulatory molecule in immune signaling and disease.
