The non-functional P2X7 receptor (nfP2X7) is a variant of the P2X7 receptor that, unlike its functional counterpart, does not form the large cytolytic pore upon activation by ATP. This prevents cell death through pore formation, allowing nfP2X7 expressing cells to evade immune-mediated killing. nfP2X7 is specifically expressed on malignant cells, making it a potential target for cancer immunotherapy, particularly CAR-T cell therapy. It is characterized by the exposure of the E200 epitope on its extracellular domain.
