NNRTIs represent a key class of antiretroviral agents that block HIV replication by binding noncompetitively to an allosteric site on the viral reverse transcriptase enzyme. This action prevents conversion of viral RNA into proviral DNA without being incorporated themselves or requiring activation via phosphorylation.
