Non-P4HA3 mRNAs with seed-region complementarity refers to a collective group of messenger RNA (mRNA) transcripts that are potential off-targets for RNA interference (RNAi) therapies designed to inhibit Prolyl 4-hydroxylase subunit alpha 3 (P4HA3). P4HA3 is a key enzyme in collagen synthesis, often overexpressed in the tumor microenvironment and associated with poor prognosis in various cancers (UniProt P59813). The specificity of RNAi-based drugs, such as siRNAs, is largely determined by the seed region, a 6-8 nucleotide sequence that guides the RNA-induced silencing complex (RISC) to its target. When transcripts other than P4HA3 share complementarity with this seed sequence, they may undergo unintended degradation or translational repression (Jackson et al., 2003, Nature Biotechnology). This phenomenon poses a significant challenge in drug development, as silencing these non-target mRNAs can lead to off-target effects and associated cellular toxicity (Birmingham et al., 2006, Nature Methods). Consequently, these mRNAs are not therapeutic targets themselves but are critical variables in the safety assessment of P4HA3-directed oligonucleotides. Identifying and minimizing interactions with these transcripts is essential for ensuring the precision and safety of gene-silencing interventions.
