Overview

Non-receptor tyrosine-protein kinase TNK1 (TNK1) is a serine/threonine and tyrosine kinase belonging to the ACK family, distinguished by its unusual C-terminal ubiquitin-association domain. TNK1 is widely expressed in fetal tissues, with selective adult expression in organs such as the prostate, testis, ovaries, colon, and intestinal epithelium. It regulates cellular growth and survival through signal transduction pathways, with roles in both negative regulation of the Ras-MAPK pathway and in mediating apoptosis, inflammation, and immune responses. TNK1 interacts with 14-3-3 proteins and is regulated by MARK-mediated phosphorylation on serine 502, which influences its localization and activity. Dysregulation of TNK1 has been linked to cancer (both as a driver and possible suppressor), inflammation, and organ injury. Pharmacological inhibition (e.g., TP-5801) demonstrates anti-proliferative effects in preclinical cancer models, but no approved therapies target TNK1 directly. Potential therapeutic targeting must consider tissue specificity and its complex involvement in cell fate decisions.

Small-molecule kinase inhibition (TP-5801 inhibits TNK1 kinase activity by blocking proliferation in TNK1-transformed cells). RNAi silencing leading to decreased cell viability and increased apoptosis (experimental context).
