Overview

Non-SMC condensin I complex subunit H (NCAPH) is a regulatory kleisin subunit of the condensin I complex, essential for the conversion of interphase chromatin into condensed mitotic chromosomes[1][2][4]. NCAPH ensures proper chromosome condensation, chromatid cohesion, and segregation, playing a key role in maintaining genomic stability and the integrity of cell division[1][2][5]. It also participates in DNA damage responses by recruiting and stabilizing specific DNA repair factors (such as GEN1), especially during DNA inter-strand crosslink repair[3][5]. NCAPH is overexpressed in a variety of cancers, promoting tumor cell proliferation, migration, and invasion, and is regarded as both a prognostic biomarker and a potential therapeutic target[2][5][6]. Loss or dysregulation of NCAPH leads to chromosome mis-segregation, cell cycle arrest, increased DNA damage, and apoptosis[3][5]. No currently approved drugs are known to target NCAPH directly as of the latest available data.
