Non-specific blood and tissue proteins at site of application refers to a broad and heterogeneous collection of proteins, such as albumin, globulins, and structural proteins like collagen, that interact with drugs at the point of administration (DrugBank, DB09153). Unlike specific therapeutic targets like receptors or enzymes, these proteins are affected through non-selective chemical or physical processes, including denaturation, precipitation, and covalent cross-linking (StatPearls, Astringents). This mechanism is primarily utilized by astringents, cauterants, and certain antiseptics to achieve localized therapeutic effects such as hemostasis, tissue destruction, or antimicrobial action (PubMed, PMID: 25607706). For instance, silver nitrate and ferric subsulfate act by precipitating these proteins to stop bleeding or treat minor skin lesions (NIH, PubChem). Because the interaction is non-discriminatory, the therapeutic index is managed by controlling the concentration and duration of exposure to avoid excessive tissue damage or necrosis (ScienceDirect, Protein Denaturation). Consequently, these agents are typically restricted to topical or local use where systemic exposure is minimized.
