Target intelligence / Profile preview
Non-specific serum protein refers to the heterogeneous group of proteins present in the blood plasma, including albumin, globulins, and fibrinogen. These proteins serve vital physiological roles such as maintaining oncotic pressure, buffering pH, and transporting endogenous substances like hormones, fatty acids, and bilirubin (StatPearls, 2023). In pharmacology, they are not typically viewed as primary therapeutic targets but are crucial determinants of drug pharmacokinetics; the extent of drug binding to these proteins dictates the concentration of free, pharmacologically active drug in the systemic circulation (PubMed, 2021). Clinical conditions that alter protein levels, such as cirrhosis or nephrotic syndrome, can significantly impact the safety and efficacy of highly protein-bound medications. Furthermore, competitive displacement between two drugs for the same binding sites on these proteins can lead to adverse drug-drug interactions (NCBI, 2022). Albumin is the primary binder for acidic drugs, while alpha-1-acid glycoprotein typically binds basic drugs, and understanding these interactions is essential for dose adjustment in specific patient populations.
Reversible non-specific binding to plasma proteins, primarily albumin and alpha-1-acid glycoprotein, which modulates the free fraction of drugs available for pharmacological action.
